$ANVS At this stage it's Cash and the Results. I think the trial is setup nicely, very nicely in fact, and I think personally we have a better than average chance at success for symptomatic relief with the trial setup. The cash situation remains tense.

A safety review by an independent monitoring board found no safety concerns and recommended the trial continue without changes. According to the company, top-line Phase 3 symptomatic efficacy data are expected after database lock and analysis, with results planned for release in 2027. Bottom line: The early results are encouraging because buntanetap showed cognitive improvement in a subgroup of Alzheimer's patients and reduced several disease-related biomarkers while appearing safe. $ANVS Anyone worrying about a reverse split because ANVS is trading in the low $1 range is way premature. A company receives a delinquency notice after its stock has an average closing price below $1.00 for 30 consecutive trading days, and ANVS has yet to close below $1 for one day. Then the NYSE gives the company at least six months (likely longer) to regain compliance. And we all know ANVS has a MAJOR readout coming 6-months from this week. Now that's worth worrying about!! 😧

$ANVS I feel like it's time for another reminder of the promising preliminary results here that suggest the ongoing Alzheimer's phase 3 will work. Both these charts are for the subgroup of mild dementia patients who test positive for "amyloid" (really, p-tau). Those are the patients who were recruited to the current Alz phase 3. On the left is 3-month ADAS-Cog results from the phase 2 Alz trial; on the right is 6-month MMSE results from the phase 3 Parkinson's trial:

$ANVS The problem is that everyone is looking at the glass as half empty. Too many Monday morning quarterbacks that are so money hungry they forget where we are at here. Let it take its course and stop trying to put the cart in front of the horse. You can’t have tomorrow until you get through today. You won’t know anything until the results come in next year, relax stop all the predictions. Maria ain’t lying, if it works it works and if it doesn’t it doesn’t. You knew that when you got involved and if you didn’t, you didn’t do your homework as a trader or investor whichever you want to be called.

$ANVS I made some good $ on this ticker back in April/May 2024. Kinda forgot about til today. I was scrolling and saw the price…did some quick research and bought 12,000 shares. With some good news(especially in March) this could easily 10X. I bought SLS, IBRX and DRTS last year and I believe by next Summer this could easily duplicate their success. I’m 100% sure I’ll get my initial investment back. Gnna buy some more if it keeps going down. Time will tell, but I’m excited to see what happens!!!

$ANVS "De-risking": ANVS: A lot of discussion has been made of the biomarker-confirmed AD population as a way to de-risk the Ph3 trial. Not so much discussion about patient over-enrollment: The original target was 760, so 862 total enrollment is +13.4%. That does NOT mean 13.4% more statistical power. Because SE falls roughly with √N, it’s about a 6.5% improvement in signal-to-noise. That matters because ADAS-Cog13 and ADCS-iADL are co-primary endpoints. For a clean Phase 3 success, you need the treatment effect to show up on both cognition AND function. IF one endpoint is sitting just above the significance threshold, that additional statistical power CAN matter. For example: ADAS p=.03 → becomes ~.02–.025 at a higher enrollment population iADL p=.055 → perhaps ~.045–.05 at a higher enrollment population

$ANVS the crash in price is not about raising money. That is no surprise. The problem is Maria does not seem to understand the clinical value of her drug. If it improves, ADS cog double the amount of that of Aricept and other generics. Patients will pay extra for the better drug. Not to mention the opportunity that the drug is producing disease modifying benefits. It is about lowering tau levels and NFL protein. If you don’t believe that, why would Biogen press forward on a compound that showed no clinical benefit. It is because it lowered tau protein levels and NFL protein levels. She’s never treated a single patient and does not understand. She desperately needs to partner with another Pharma company. If she doesn’t understand how to make this happen then she needs to hire a clinical person that’s not retired and actually treats patients with Alzheimer’s disease to help her understand her compound. I was extremely disappointed in her performance. She needs help.

I have a large position, but we need to do something about Maria's complete ineptitude from a basic corporate governance and stakeholder communication perspective. My proposal is that we draft a simple paragraph calling for a new CEO, for her to step aside to COO, list everyone's name and the number of shares each name represents, and then send it to the Board. I'm happy to organize this. Like this post if you are willing to participate.

$ANVS Bottom Fishing here. Ironically, this company is located right down the street from where I currently live. They are right next to Vanguard Corporate Headquarters. R.I.P. Jack Bogle. No one has this thing cheaper than $1.04. Dilution and obvious concern, but 18 million might keep them rolling for a little bit with no debt. Never had a R/S either.

$ANVS Next big catalyst starting next week is the AD OLE study for patients completing 18-month FDA AD trial. ANVS team will not know which patient is a placebo or took the drug, but biomarker data collected during AD OLE onboarding and patient participation will be something to keep an eye out.

BUT IF YOU THINK THAT PART OF ANY DEAL AT THIS POINT AND THE CHANCE THIS DRUG IS A SUCCESS AND BECOMES A FRANCHISE DRUG THAT MARIA WOULD CHANCE A BUYOUT WITH HER RELINQUISHING ANY RIGHTS SHE HAS TO THE LONG HARD WORK SHE'S PUT INTO MAKING IT A SUCCESS, YOU DON'T KNOW MARIA MACCECCHINI. A CONGLOMERATE PHARMACEUTICAL COMPANY AS PART OF A DEAL WOULD WANT HER TOTALLY OUT OF THE PICTURE AND THAT WILL NEVER HAPPEN UNTIL SHE SAYS SO.

$ANVS As stated by Maria, the six-month P3 symptomatic AD trial ends in December 2026, and 6-months-ish from now (during mid-March 2027) data readout occurs. If results are positive, ANVS market cap will respond dramatically and raising capital at minimal share prices will end as just about everyone will want a piece. So, ANVS needs enough dollars to survive for six months, of which they may have enough to survive half of that now. Dilution is always a pain in the ass, especially at such low prices, but IF BUNTANETAP WORKS the upside remains monstrous! IMO, the Webinar lowballed the value of success, which was likely deliberate as it's always better to overperform in the end. Currently, there is no drug making a substantial and sustained difference in symptoms or disease modification for ANY dementia related illness. IF buntanetap is that drug...I need say no more! ⬆️⬆️ I increased my position today by 12% and I'm not finished!!

$ANVS I have a very different impression of the webinar than - I don't think Maria was discounting the effect of the drug or the value of a symptomatic treatment for Alz at all. I think the confusion may arise from Maria saying she does not plan to commercialize buntanetap for symptomatic use, even though she will apply to the FDA for it. That is a practical matter stemming from the timing - FDA approval for symptomatic treatment would occur around the same time as the 18-month disease-modifying results. If buntanetap is disease modifying, then it's not worth marketing it very temporarily as symptomatic. Aside from that practical matter, I also think that's when Maria will be ready to sell the company...
